Alnylam Pharmaceuticals Inc. (ALNY) is offering shareholders a study in contrasts. The biotech has rebounded roughly 25% since the end of July, yet remains down 35% year-to-date. Wall Street, for now, is unfazed: of the 21 analysts covering the stock over the past three months, 18 rate it “buy” or “strong buy,” and the consensus 12-month price target implies about 44% upside.
The bull case starts with valuation. Alnylam trades at just 19.7 times forward earnings — cheap for a growth stock by conventional yardsticks. Its price-to-earnings-to-growth ratio, based on analysts’ five-year earnings estimates, sits at a scant 0.32.
The competitive backdrop has also tilted in the company’s favor this year. Pfizer Inc.’s (PFE) Vyndamax franchise is now insulated from generic competition until mid-2031, clearing a potential near-term cloud over Alnylam’s Amvuttra. More decisively, AstraZeneca Plc (AZN) and Ionis Pharmaceuticals Inc. (IONS) said in July 2026 that Wainua (eplontersen) failed its primary endpoint in a late-stage trial in transthyretin amyloid cardiomyopathy (ATTR-CM). A prospective rival has, in effect, exited the frame — a development that has lifted management’s confidence in nucresiran, its own Phase 3 candidate being studied in both ATTR-CM and hereditary ATTR amyloidosis with polyneuropathy (ATTR-PN). Much of Alnylam’s broader pipeline is wholly owned or company-led.
Yet the narrative has cracks. Alnylam has already cut its full-year revenue guidance, after early demand growth for Amvuttra in ATTR-CM “benefited significantly from pent-up demand for a new therapy that has since normalized.” Should third- and fourth-quarter 2026 results fail to show that normalization was only temporary, some analyst downgrades would hardly be surprising.
A second wildcard remains: AstraZeneca and Ionis could still release further Wainua trial details that rekindle concerns around nucresiran.
Absent negative developments on either front, Wall Street’s bullish stance on Alnylam looks defensible, and the roughly 44% consensus upside appears within reach. The real showdown between bulls and bears will play out in the next two quarters of Amvuttra demand data.
