U.S. biotech Immunome (NASDAQ: IMNM) reached a major milestone this week after announcing overwhelmingly positive Phase 3 results for its lead candidate varegacestat in treating desmoid tumors—a rare and locally aggressive form of cancer. The news propelled the company’s stock to close up 15.69% at $22.64 on Monday, reflecting strong investor confidence in this breakthrough therapy.
The registrational trial met all primary and key secondary endpoints. Data revealed that varegacestat reduced the risk of disease progression or death by 84% compared to placebo, with statistically significant improvement in progression-free survival.
The drug also demonstrated impressive tumor shrinkage and symptom relief:
“Desmoid tumors place a significant physical and emotional burden on patients due to their unpredictable behavior and the limited treatment options available,” said Dr. Mrinal Gounder, the trial’s primary investigator. “These results represent a crucial step forward in the clinical management of this disease.”
Varegacestat was generally well-tolerated, with common adverse events including diarrhea, fatigue, rash, nausea, and cough. Notably, comparative analyses suggest a potential safety advantage over the currently approved therapy Ogsiveo (from Merck KGaA), particularly regarding ovarian toxicity rates.
Analysts were quick to highlight the drug’s competitive edge. Wedbush analyst David Nierengarten described the outcome as a “knockout” and stated that varegacestat “outperforms the benchmark set by Ogsiveo,” positioning it to become a best-in-class therapy for desmoid tumors.
Based on these compelling results, Immunome plans to submit a New Drug Application (NDA) to the U.S. FDA in the second quarter of 2026. “These findings underscore the potential of varegacestat to deliver best-in-class efficacy through a convenient once-daily oral regimen, helping patients reclaim their lives,” said Clay Siegall, CEO of Immunome.
The commercial launch strategy is already taking shape. Analysts expect the company to initially focus on approximately 85 specialized sarcoma centers across the United States, which treat the majority of desmoid tumor patients. Payor coverage at launch is anticipated to exceed 90%.
The positive clinical data not only triggered a sharp rise in Immunome’s share price but also reinforced its strong standing in the biotech investment landscape. As varegacestat advances steadily toward regulatory submission and potential approval, Immunome is poised to redefine the treatment paradigm for desmoid tumors while presenting a compelling growth opportunity for investors.
